RP-HPLC analysis of the displacement of AA from Eallo of huPGHS-2 by unlabeled DHA, DPA, adrenic acid, linoleic acid, or PA

[1-14C]AA (1 μm) was incubated with huPGHS-2 (1 μm) at 37 °C for 4 min and then the indicated amounts of an unlabeled FA was added, and the incubation was continued for another 4 min. The reactions were stopped by the addition of ethyl acetate/acetic acid (20:1), and an aliquot of the organic phase was subjected to radio-reverse phase-HPLC to separate the radioactive products and unreacted AA as described under “Experimental Procedures.” The results are shown as the percentage of total 14C label that remained in the RP-HPLC fraction co-eluting with unreacted AA.
