Simultaneous whole-cell recordings of pre-and postsynaptic currents from two adjacent Chx10-Puro cells 31 days after puromycin selection. Voltage steps to 0 mV from a holding potential of −80 mV elicited inward sodium current, and uncompensated capacity transients, in the presynaptic cell. Excitatory synaptic currents (EPSCs) were evoked in the postsynaptic cell with a delay of 3.2 msec, measured from the peak of presynaptic sodium current to the initial rise of the EPSC. Exposure to the AMPA/kainate receptor antagonist NBQX (30 μM) completely blocked the EPSC, while the GABAA receptor antagonist bicuculline methiodide (0.2 mM) had no effect.
