Model for limitation of Cmr nuclease activity to invader DNA. (A) The Cmr complex DNA nuclease is activated and tethered by the nascent target RNA. The transcribed invader RNA containing the crRNA target sequence (orange) and rPAM (red) is recognized by the Cmr complex containing the crRNA with the guide sequence (gray). The activated nuclease is tethered to and cleaves the invader DNA. (B) As the length of the tether increases, the Cmr complex DNA nuclease is deactivated by cleavage of the target RNA.
