(A) Thrombin (Th)-catalyzed fibrin polymerization was monitored at the indicated concentrations of compound 1 (white box), 2 (light gray box), 3 (dark gray box), and 4 (black box) using a catalytic assay, as described in the “Materials and Methods” section. The results are expressed as the Vmax values as percentage of the controls. (B) The effect of each compound on human platelet aggregation induced by 2 mM U46619. (C) Each compound in DMSO was injected intravenously at the indicated concentration. The effects of each compound on mouse platelet aggregation induced by 2 μM U46619 (U) were monitored ex vivo. (D) The effect of each compound on cellular viability was measured using the MTT assay. D = 0.2% DMSO used as the vehicle control. The data represent the means ± SEM of three independent experiments performed in triplicate. *p < 0.05 vs. Th (A) or U46619 (B,C) alone.
