WT or serglycin−/− PCMCs were activated with IgE‐receptor crosslinking (IgE/Ag) or calcium ionophore A23187 (CI). Supernatants were collected after 1 h and analyzed for chymase‐, tryptase‐, and CPA‐like activities by adding fluorogenic or chromogenic substrates. Experiments were performed in triplicates.
Activities are given as mean ± SEM of raw fluorescence units (RFU)/(min × 106 cells).
Activities are given as mean ± SEM of ΔmOD/(min × 106 cells). Note that hydrolysis of the CPA substrate results in a decreased absorbance.
