Interaction between Mediator complex subunits of Arabidopsis. (A) Yeast two-hybrid assay of the interaction between AtMed4 and AtMed9. Yeast cells expressing AtMed4 with AD vector alone and AtMed9 were spotted and grown on SD-Trp/Leu/Ade media. (B) AtMed4 and AtMed9 cDNAs were cloned in BiFc vectors and bombarded on onion peel. Fluorescence was observed under confocal microscope after overnight incubation of the peel at 22° C. DAPI (blue) staining was used to identify nuclei in the cells. (C) Interaction was fine mapped using deletion constructs of AtMed4 andAtMed9. Upper panel shows yeast two-hybrid assay whereas lower panel shows line diagram of IDRs in the deletion constructs. Red boxes represent IDR regions. (D) Schematic view of AtMed19 deletion constructs used in yeast two-hybrid experiments. Red regions represent IDRs.
