Figure 6. — Interaction between Mediator complex subunits of Arabidopsis. (A) Yeast two-hybrid assay of the interaction between AtMed4 and AtMed9. Yeast cells expressing AtMed4 with AD vector alone and AtMed9 were spotted and grown on SD-Trp⁻/Leu⁻/Ade⁻ media. (B) AtMed4 and AtMed9 cDNAs were cloned in BiFc vectors and bombarded on onion peel. Fluorescence was observed under confocal microscope after overnight incubation of the peel at 22° C. DAPI (blue) staining was used to identify nuclei in the cells. (C) Interaction was fine mapped using deletion constructs of AtMed4 andAtMed9. Upper panel shows yeast two-hybrid assay whereas lower panel shows line diagram of IDRs in the deletion constructs.
