CD8+ T cells from B6.CD40+/+.Thy1.1 and B6.CD40−/− CD45.2+ mice were mixed at 1:1 ratio and intravenously injected into congenic B6.CD45.1.Thy1.2 mice (8 × 106 of each cell type/mouse) followed by BALB/c heart allograft placement and mATG treatment. A. The percentages of tracer CD8 T cell populations (Thy1.1+CD45.2+ WT and Thy1.1CD45.2+ CD40−/−) within total live peripheral blood cells were determined by flow cytometry and presented as mean ± SEM, N = 4 mice per group. B. The numbers of transferred CD8 T cells in the spleens of individual recipients and group means were determined on day 63 after mATG treatment.
