The DNA products from RT-PCR analysis are shown after restriction enzyme digestion, electrophoresis and visualization with fluorescent dye. Purified mRNA transcripts from P. sojae parental strains and test cross progeny with the genotype Avr3a^P6497/Avr3a^P7076 were analyzed to determine allele specific expression. The RT-PCR was performed using Avr3a specific primers and the amplified product was digested with the restriction enzyme AluI. The Avr3a^P6497allele contains a restriction site for AluI whereas the Avr3a^P7076 allele does not. P. sojae strains are indicated above each lane; M, marker lane.
