Fig. 4 — Optimization of conditions for UV binding of THPs to collagen films. Biotinylated THPs were added to wells coated with 100% EDC/NHS cross-linked collagen films. Following PBS and citrate buffer washings, attached peptides were detected through HRP-Streptavidin binding to Biotin. A) Diazirine-ES-GFOGER-Biotin was incubated at concentrations ranging from 0 to 10 μg/ml and exposed to long wavelength UV light for 5 min. B) Diazirine-ES-GFOGER-Biotin or ES-GFOGER-Biotin were added to collagen films at 5 μg/ml and exposed to long wavelength UV light for durations ranging from 0 to 60 min.
