Total RNA was isolated and the exsA mRNA levels were determined by real time PCR with proC as an internal control. Bacteria were grown to an OD₆₀₀ around 1.0 in LB with or without 5 mM EGTA. Intracellular and secreted ExoS were separated by centrifugation. Samples from equivalent bacterial cells were loaded into SDS-PAGE gels and probed with antibody against ExoS.
