The hypersensitivity of ERAD mutants to sr7575 is conserved in A. fumigatus. (A) Conidia from A. fumigatus WT and deletion mutants were recovered in 0.05% Tween–water, and serial dilutions were spotted onto sr7575-containing RPMI 1640, pH 7.0. Plates were incubated at 37°C for 72 h. (B) Analysis of UPR target gene expression (bipA and tigA) by qRT-PCR. Cultures were treated with sr7575, DTT, or sr7575 for 1 h followed by DTT. RNA was extracted and analyzed by qRT-PCR, using tubA mRNA for normalization. The results of treated versus untreated samples from three independent experiments are shown.
