S1PR1 expression controls endogenous effector T cell behavior at the lymphatic border. GzmBER^T2/ROSAEYFP (GzmB YFP) or GzmBER^T2/ROSAEYFP-S1P1^flx/flx (GzmB YFP S1PR1^fl/fl) mice were infected with VSV in the footpad and treated orally with tamoxifen starting at day 3 p.i. At day 6 p.i., the dLN was surgically exposed for intravital 2P imaging. Twelve hours before imaging, fluorescently conjugated anti-LYVE-1 antibody was injected into the footpad to highlight the lymphatic vessels in the dLN. Movies of effector T cells interacting with the LYVE-1 region were analyzed on the software Imaris.
