At DIV4, inhibition of ribosomal biogenesis affects global protein synthesis. A and B, DIV1 hippocampal neurons were transfected as described for Fig. 10. On DIV4, cells were treated with 50 ng/ml BDNF for 45 min as indicated. Then OPP was added for an additional 30 min followed by fixation and visualization of nascent peptides as in Fig. 7, A and B. A, representative images of transfected neurons (arrows). B, in shS6-, shL4-, or shTIF1A-transfected cells, nascent peptide accumulation was significantly decreased. BDNF was unable to rescue such a deficit.
