Effector binding and orientation states. Superposition of representative structures from (A) OS1 and (B) OS2 indicates no steric clash and substantial overlap between Raf (green) and bilayer (ice blue), respectively. An orientation where neither lobe 1 nor lobe 2 is in close proximity with membrane is capable of interacting with effector (C), while an intermediate orientation that is similar to OS2 results in partial overlap of the effector with the membrane and therefore may not be favored (D).
