A, Co-localization of Smad2 and M10 in lung fibroblasts cultured in serum free medium without TGFβ (panels 1, 2, and 3) and with TGFβ (panels 4, 5, and 6). B, M10 reduces TGFβ-induced Smad2 phosphorylation in normal lung and skin fibroblasts. Cells were cultured on 6-well plates to 90% confluence, serum-starved overnight, incubated with or without M10 or scrambled peptide (Scr) and TGFβ, and subjected to immunoblotting as detailed in the Methods section.
