Related to Fig. 2. Pre- and postsynaptic expression of 5-HT2A receptors in the PFC. (A) The region corresponding to the dotted box was dissected from cortical slices and used to prepare the synaptosomal fraction. Synaptosomes were treated with 1% TX-100 containing buffer, and equal quantity of proteins from soluble (presynapse) and insoluble (postsynapse) fractions were analyzed by Western blotting using the indicated antibodies. As expected, PSD-95 was only detected in the postsynaptic fraction, whereas synaptophysin and Munc-18 were exclusively found in the presynaptic fraction. (B) Specificity of the 5-HT2A antibody. Synaptosomes from WT and 5-HT2A−/− mice were analyzed by Western blotting using 5-HT2A receptor and actin antibodies.
