The ectopic expression of both AtYchF1 and AtYchF1-G4′ reduced the tolerance toward salinity stress. The same transgenic lines as described in Fig. S6 were used in this study. Phenotypes of leaf chlorosis (A) and chlorophyll contents (B) were examined by using 10-d-old AtYchF1- or AtYchF1-G4′-transgenic Arabidopsis seedlings further grown on MS agar medium supplemented with 150 mM NaCl for 7 d. Transgenic lines overexpressing AtYchF1 or AtYchF1-G4′ showed significantly reduced salt tolerance. There were no significant difference (one-way ANOVA, P < 0.05) between AtYchF1 and AtYchF1-G4′ transgenic lines. (C) Expressions of abiotic stress responsive genes rd22 and rd29a were observed 1 d after salt treatment by using real-time PCR (Table S2). All experiments were performed twice, and similar results were obtained.
