Figure 4.

Canonical TRP channels contribute to store-operated signals in mouse Müller glia. A, The peak amplitude of the depletion-evoked Ca2+ response in Trpc1/3−/− cells (n = 14, N = 4) is reduced compared with that of WT controls (n = 16, N = 3). The SOC channel-inhibitor cocktail evokes a significant (p < 0.05) additional reduction in peak SOCE (n = 14, N = 4). B, Representative I–V curves taken during peak SOCE in 2 mm [Ca2+]i. C, The selective TRPV3 antagonist Pyr3 (5 μm) has no effect on the depletion-induced increase in fura-2 ratio. D, Summary graph for the Trpc1/3−/− cohort in the presence/absence of the inhibitor cocktail. E, TRPC channels disproportionately contribute to high-amplitude SOCE. Distribution of peak overshoot amplitudes in wild-type, Trpc1/3−/−, and cocktail-treated cells.
