The effect of PEG surface coatings on the behavior of biodegradable PBAE-based DNA-NPs in vitro, ex vivo and in vivo. (A) PEG-coated PBAE-MPP remained stable in BALF at 37ºC, whereas uncoated PBAE-CP rapidly aggregated. (B) PBAE-MPP diffused rapidly in freshly expectorated CF sputum, whereas PBAE-CP were largely hindered or immobilized. Immobilization in CF sputum correlated with poor distribution of (C) PBAE-CPs in the airways of mice in vivo, whereas (D) PBAE-MPPs distributed more uniformly in the mouse trachea.
