Fluorescein (FITC) conjugated to the minimal peptide Ybbr was subjected to PPTase activity evaluation using standard PPTase reaction conditions, including rhodamine-CoA. Reduction in fluorescent signal for fluorescein indicates rhodamine-pantetheine conjugation, and thus PPTase activity. Human PPTase (hPPTase) possesses poor labeling activity compared to Sfp under these conditions.
