Inhibition of calcium signaling decreases cisplatin-induced mitochondria-mediated apoptosis in HeLa cells. (A) HeLa cells were treated with cisplatin (5 µg/ml) with or without BAPTA/AM (2.5 µM) and 2-APB (100 µM) for 24 h and then stained with Annexin-V. Data are presented as the mean ± SD (n=3). (B) HeLa cells were treated with cisplatin (5 µg/ml) with or without BAPTA/AM (2.5 µM) and 2-APB (100 µM) for 24 h, and stained with Hoechst 33342. Cell morphology was observed by confocal microscopy (scale bar, 30 µm). (C) The expression of calpain, caspase-3 and cleaved caspase-3 in HeLa cells treated with cisplatin (5 µg/ml) with or without BAPTA/AM (2.5 µM) and 2-APB (100 µM) for 12 h by western blotting.
