Rab11 plays an important role for WNV replication. A, Rab11 expression levels in Rab11 KD cells. WT or Rab8b KO MEFs were infected with recombinant lentivirus carrying shRNA against mouse Rab11a expression cassette (Rab11a KD). shRNA transduced MEFs were transfected with siRNA against mouse Rab11b (Rab11a+b KD). These cells were analyzed by immunoblotting for Rab11 and actin. B, viral titer in culture supernatants of WT, Rab11a+b KD WT, Rab8b KO, and Rab11a+b KD Rab8b KO MEFs. After 24 h, siRNA-transfected cells were infected with WNV (1 Pfu/cell), culture supernatants were harvested at 48 hpi, and viral titers were measured by plaque assay.
