The orphan nuclear estrogen-related receptor α (ERRα) is an early target of endogenous, neuronal PGC-1α. (a) Silencing of neuronal PGC-1α repressed the expression of endogenous ERRα by more than 90%, respectively, compared to controls transfected with scrambled siRNAs (NS). Model circuit and observed effects in circuit components are shown. (b) Silencing of ERRα largely recapitulated the reduction in electron transport chain gene expression observed in response to PGC-1α-silencing (note log2 scale). NRF1 gene expression was downregulated by more than 50% by silencing PGC-1α (c) or ERRα (d). The ribosomal gene RPL13 was used as control for input RNA.
