(A) HEK 293T cells were co-transfected with an AP-1-driven luciferase reporter and the indicated siRNA of TRIM69. Relative luciferase activity was measured and statistically analyzed by unpaired t-test, p < 0.01; (B) HEK 293T cells were co-transfected with signal-pathway-driven luciferase reporter plasmids and Flag-TRIM69. Relative luciferase activity was measured and statistically analyzed by unpaired t-test, p < 0.01; (C) HEK 293T cells were transiently transfected with different concentrations of Flag-TRIM69 (0.2, 0.4, and 0.8), AP-1-driven luciferase reporter and c-Jun. Relative luciferase activity was measured and statistically analyzed by unpaired t-test, p < 0.05; (D) HEK 293T cells were transiently transfected with different concentrations of Flag-TRIM69 (0.2, 0.4, and 0.8) and AP-1-driven luciferase reporter. Cells were treated with 10 ng/ml TNFα for 8–12 h. Relative luciferase activity was measured and statistically analyzed by unpaired t-test, p < 0.05.
