(A–B) OVCAR-3 cells were incubated for 48 h in the presence of 30 nM FK866, with (grey bars) or without (white bars) 5 μM dipyridamole (dyp) and NMN or NR (added twice/day at 10 μM final concentration), as indicated. NAD+ content (A) and cell viability (B) were evaluated after 48 h. Cell viability was expressed as relative to control, untreated cells. (C, D) OVCAR-3 cells were transfected in the presence of a specific NRK1-siRNA (black bars) or negative control-siRNA (white bars). Twenty four hours after transfection, cells were treated (or not) with 30 nM FK866, in the presence or absence of NMN or NR (added twice/day at 10 μM final concentration): NAD+ content (C) and cell viability (D) were evaluated after 48 h and 72 h, respectively. Results are the mean ± SD of at least 3 independent determinations. *p < 0.05.
