FIGURE 7. — Subcellular localization of full-length DGKη and its mutants (DGKη-PH+C1D, DGKη-PH, and DGKη-C1D) in COS-7 cells treated and untreated with hyperosmotic shock. AcGFP alone (A), AcGFP-tagged full-length DGKη1 (splice variant 1) (B) and its mutants DGKη-PH+C1D (C), DGKη-PH (D), and DGKη-C1D (E) were co-expressed with DsRed monomer-PLCδ1-PH in COS-7 cells. DsRed monomer alone (F) and AcGFP-tagged DGKη-PH (G) were co-expressed with AcGFP-PLCδ1-PH in COS-7 cells. After 48 h of transfection, the cells were incubated with 500 mm sorbitol in DMEM for 30 min. The cells were fixed with 3.7% formaldehyde and imaged by inverted confocal laser microscopy (FV1000-D). Representative data of three independent experiments are shown.
