Figure 3.

Positional cloning of the WP1 gene. A, Map-based cloning of the WP1 gene. The WP1 locus was initially mapped to the short arm of chromosome 7 between two simple sequence repeat markers, Bs8 and Bs1. The WP1 locus was narrowed to a 43-kb region between Bs12 and Bs17 on BAC clone P0428D12 using 958 homozygous mutant plants. B, Structure of WP1. ATG and TGA represent the start and stop codons, respectively. Black boxes indicate the exons and the lines between them indicate introns. A single nucleotide replacement led to an amino acid change. C, Verification of the change between wild-type and wp1 genomic DNAs with the derived cleaved amplified polymorphic sequence marker jc1. D, Functional complementation of WP1: Seedlings of transformation experiments. E, Panicle phenotypes of the wild-type, wp1w, and three independent transgenic complementation lines.
