A. PC3 and DU145 cells were treated with PA (20μM) with or without compound C (20μM), metformin (10mM) and metformin (10mM) with PA (20μM) for 24h. Cell lysates were then examined for mTOR signaling downstream of AMPK using western blotting. B. Cells lysates of PC3 and DU145 were analyzed for expression of autophagy marker (LC3AB) and apoptosis marker (caspase 3) after treating with PA alone or in combination with compound C and metformin. C. Cells were labeled with FITC and analyzed by flow cytometry. Percentage of apoptotic cells with the corresponding dose of PA or CC is shown on the histograms. The experiment was performed in triplicates and repeated thrice.
