(A) SKBR3 cells were co-transfected with the CSL luciferase reporter and scrambled control siRNA or Notch-1 siRNA for 24 h and then treated with RY10-4 or vehicle control. (B) Representative micrographs of the cells with 100 nM Notch-1 siRNA treatment, 1 μM RY10-4 treatment, and combinational treatment for 24 h. (C) Methylene blue was used to evaluate the cell viability following different treatments. (D) Western blot analysis detected Notch-1 and β-actin proteins in SKBR3 cells previously transfected with scrambled siRNA or Notch-1 siRNA for 24 h.
