(a) HAEC were pretreated with Mito-Esc (2.5 μM) for 2 h prior to the addition of H2O2 (0.5 mM) for 8 h and mitochondria genome-encoded gene expressions as indicated were measured by qRT-PCR. (b) HAEC were treated with various concentrations of H2O2 (0.25–0.75 mM) for 8 h and PGC-1α, SIRT3 and TFAM mRNA levels were measured by RT-PCR. (c) Same as b except that, protein levels were measured by Western blot analysis. (d) Same as c except that cells were incubated with H2O2 (0.5 mM) for various time points (2–8 h). (e) Same as d except that, cells were treated with different concentrations of Mito-Esc (1–5 μM) for 8 h.
