(ac) Cells were treated with DMSO or MLN4924 at indicated concentrations for indicated periods of time (a, 24 hrs, c, 48 hrs) before subjected to Western blot analysis using antibodies against indicated proteins. (dg) Cells were transfected with siRNA oligonucleotides targeting CDT1 (d,f), or targeting p21 (e,g), along with scrambled control. Twenty-four hrs post transfection, cells were treated with MLN4924 for 24 hrs (d,f) or 48 hr (e,g). Cells were then split: one portion of cells was subjected for Western blotting analysis, using indicated Abs (d,e) and the other portion was for FACS analysis (f, left) or SA-β Gal staining (g, left).
