(AC) Livers were collected from normal individuals or patients with alcoholic steatohepatitis (n = 7). mRNA levels were determined by qRT-PCR (A). Protein levels were determined by Western blotting (B) and then quantified (C). (D,E) C57BL/6 mice were fed a Lieber-DeCarli diet for 5 days, followed by fed a Lieber-DeCarli diet containing 5% ethanol or pair-fed a control diet for 10 days. On the 16th day, mice were gavaged with a single dose of ethanol (5 g/kg) (n = 8). mRNA levels were then determined by qRT-PCR (D) and protein levels determined by Western blots (E). (F,G) Mouse primary hepatocytes were treated with 0, 50 or 100 mM ethanol for 24 h (n = 3). mRNA (F) and protein (G) levels were determined. *p < 0.05, **p < 0.01. Unpaired Student t-test was used for statistical analysis.
