CD4 and CD8 staining in spleen sections. Spleens were fixed for 24 h for each condition. Single stains with CD3, CD4, or CD8 Ab were performed, and tissue sections were imaged at 4× with a PerkinElmer Vectra platform. Spleens from Rag1-knockout mice were stained for CD3 (A), CD8 (E), or CD4 (I). Stains were also performed on spleens from pmel TCR-transgenic (B, F, and J) or TRP1 TCR-transgenic (C, G, and K) mice, both on a Rag1-knockout background. Stains were also performed on CD4-knockout mice (D, H, and L). (M) Cells were enumerated as the percentage of positive cells in a 20× field.
