Fig. 4 — The correlation between the efficacy of Zt/g4-DM1 and the number of RON receptors expressed by cancer cells. The IC₅₀ values for cell viability and cell death were calculated using the GraphPad Prism 6 software and then plotted with individual BC and NSCLC cell lines expressing different levels of RON. Colon cancer DLD-1 cells expressing low levels of RON [25] also were included for comparison. The cell surface RON numbers per cell are: DLD-1, 4480; MDA-MB-231, ~8185; H228, 10,207; DU4475, 14,841; H358, 15,286; and T-47D, 15,756 RON receptors per cell. The IC₅₀ values at or less than 5 μg/ml of Zt/g4-DM1 were used as the effective dose to determine the RON receptor number required to reach the IC₉₅ value. Results shown here are from one of three experiments with similar results
