Comparison of ligation under the SAGE protocol versus under directed ligation chemistry. Ligation reactions were performed in 5% PEG-8000 (w/v) in the presence or absence of NlaIII, using standard SAGE adaptors (adaptor 1) or modified SAGE adaptors with an N6-methyl-deoxy-adenosine base (adaptor 1m6A) at a final concentration of 1 μM. The reactions were performed at a final reaction volume of 10 μl and incubated as described in Materials and Methods.
