Mobility of eGFP-H2B during preimplantation development Complementary RNA (cRNA) encoding eGFP-tagged H2B (eGFP-H2B) was injected into the cytoplasm of 1-cell stage embryos 2 h post-insemination (hpi). The mobility of eGFP-H2B was analyzed using FRAP in embryos at the 1-, 2-, 4-, morula, and blastocyst stages during 10–13, 28–32, 45–48, 70–72, and 94–98 hpi, respectively. The recovery curve and mobile fraction are shown on the top and bottom, respectively. In the bar graph of the mobile fraction, different characters indicate statistical differences among developmental stages (P<0.05; by Student t-test) and the error bar indicates the standard error. More than 3 independent experiments were performed for each developmental stage and the data were accumulated. In total, at least 23 or more nuclei were analyzed at each developmental stage.
