Fig 5. L. acidophilus K301 induced ABCA1 and ABCG1 production through the endogenous LXR agonist, 24(S), 25-epoxcycholesterol.

Fig 5

(A) PMA-differentiated macrophages were incubated with TO901317 and heat-killed L. acidophilus K301 in medium containing [14C]-acetate for 16 h. Neutral lipid extracts were separated by TLC and visualized by exposure to X-ray film. (B) Heat-killed L. acidophilus K301-stimulated THP-1 macrophages were treated with a specific inhibitor of endogenous ligand synthesis, RO 488071, and neutral lipid extracts were separated using TLC. (C) Heat-killed L. acidophilus K301-stimulated THP-1 macrophages were treated with RO 488071 and mRNA levels of ABCA1 and ABCG1 were determined using real-time PCR. mRNA levels were normalized to GAPDH. Data are given as means ±SD. **P < 0.01 vs. untreated cells. The error bars in individual samples are shown as variations from triplicate assays.
