A) YAMC cultures were transfected with vehicle control or PTK6 siRNA then treated with vehicle control or TNFα/IFNγ for 16 hours. Nuclear fractions were harvested then assayed for presence of FoxO1. PTK6 was assayed in whole cell lysate to ensure knockdown. Densitometric calculations were based on 3 separate experiments (please see S4 Fig for full blot scan). p<0.05, n = 3. B, C) Either PTK6-/- or PTK6 +/+ were grown to confluence then treated with TNFα/IFNγ for 24 hours as indicated in Methods. Whole cell lysates were assayed by Western blot (please see S4 Fig for full blot scan). Results are representative of 3 separate experiments.
