Protoplasts show that βCA2 and βCA4.2 are located in the cytoplasm and βCA4.1 is located in the plasma membrane. Confocal images of leaf cell protoplasts generated from COL, βCA2-eGFP, βCA4.1-eGFP, and βCA4.2-eGFP leaves confirm the presence of βCA2 and βCA4.2 in the cytosol and βCA4.1 in the plasma membrane. Wild-type (COL) leaves were used as a negative control.
