(a) RhoC-GTP levels were increased in EC after Thrombin (1 U/ml) stimulation, as analyzed using a Rhotekin-RBD pull down. Total RhoC levels were detected to control for equal RhoC levels and loading. (b) EC were grown to a monolayer on electrode arrays. Resistance of the endothelial monolayer was measured on the ECIS before and after stimulation of thrombin at t = 0:41 hours. (c) EC were stained for RhoC, Actin and VE-cadherin and were followed over time after Thrombin stimulation (1 U/ml). Arrowheads indicate RhoC-specific cell-cell contact areas where RhoC is present in unstimulated EC, disappears at Thrombin = 10, 30 min and relocates at Thrombin = 60, 120 min. Bar = 25 μm.
