Interferon (IFN)β inhibits the upregulation of adhesion molecules in vivo and in vitro. A, RNA was extracted 24 hours after reperfusion from sham control, vehicle‐, and IFNβ‐treated middle cerebral artery occlusion/reperfusion (MCAO/R) mice (n=7/group) and analyzed for expression of E‐selectin and ICAM‐1 by quantitative polymerase chain reaction (Q‐PCR). Data are presented as the group mean±SEM. B through D, bEnd.3 cells were pretreated with IFNβ with different concentration (10 to 1000 U/mL) for 1 hour followed by tumor necrosis factor (TNF)α (50 ng/mL) for 1 hour. mRNA expression of E‐selectin and ICAM‐1 was determined by Q‐PCR (B). bEnd.3 cells were pretreated with IFNβ (1000 U/mL) for 1 hour followed by TNFα (50 ng/mL) for 4 hours. The surface expression of ICAM‐1, E‐selectin, and P‐selectin was determined by FACS (C and D).
