VP8 prevents IFN-β-induced nuclear accumulation of STAT1. Vero cells were transfected with pFLAG (A), pFLAG-VP8 219-741 (B), or pFLAG-VP8 (C). At 24 h posttransfection, cells were stimulated with 2,500 units of human IFN-β for 30 min or left untreated, followed by fixation with paraformaldehyde and permeabilization. Truncated and full-length VP8 proteins were incubated with monoclonal anti-FLAG antibody and Alexa Fluor 488-conjugated goat anti-mouse IgG. STAT1 was detected with rabbit anti-STAT1 antibody and Alexa Fluor 633-conjugated anti-rabbit IgG. The nucleus was stained with Prolong gold DAPI. The cells were examined with a Leica SP5 confocal microscope.
