Diminished mitochondrial function in the primary embryonic fibroblast cells from the MnSOD-deficient mice. (a) Superoxide radicals produced in the cells were analyzed using a DHE fluorescence probe. Antimycin was added to induce cellular superoxide radicals and PEG-SOD was used to catalyze the produced superoxide radicals. (b) Mitochondrial respiration was analyzed by quantification of the oxygen consumption rate (OCR). Rotenone was used to fully inhibit oxygen consumption. (c) Cellular glycolysis was analyzed by estimating the extracellular acidification rate. 2-Deoxy-D-glucose was added to inhibit the cellular glycolytic pathway.
