RE1, RE2, RE3, and RE4 cooperatively prevent Bcl10 association with CARD11. A–C, HEK293T cells were transfected with expression vectors for the indicated Myc-tagged CARD11 variants and FLAG-Bcl10 as indicated. Anti-FLAG immunoprecipitations (IP) were performed as described under “Experimental Procedures” and analyzed by Western blotting (WB) with the indicated primary antibodies. The presence of a mutated RE in a construct is indicated by blue lowercase letters (e.g. re1), and the presence of a wild-type RE is indicated by red capital letters (e.g. RE1). D, Jurkat T cells were retrovirally transduced to express the indicated Myc-tagged CARD11 variants.
