Reactions containing 4.50 mm CMP-Neu5Ac (donor), 3.0 mm acceptor (Gal-β-1,3-GalNAc-α-O-Bn, lactose or N-acetyllactosamine) and sialyltransferases at a final concentration of 1.0 or 2.0 μm where prepared in 20.0 mM MOPS, pH 7.5 in 40.0 μL final volumes. Reactions were incubated at 37°C and samples analyzed over the reaction time of 22 h. Progress in the synthesis of 3’-sialyl-Gal-β-1,3-GalNAc-α-O-Bn (A), 6’- sialyl-N-acetyllactosamine (B) and 6’- sialyllactosamine was monitored by HPAEC-PAC.
