Anti-mouse CD47 nanobody A4 potently antagonizes the CD47–SIRPα interaction across a range of mouse but not human tumor cell lines. (A) Dose–response curves demonstrating inhibition of SIRPa binding to cell surface CD47 on mouse LSTRA, CT26, Tubo-EGFR (A), and human Raji (B) tumor cells with the anti-mouse CD47 nanobody A4 or CV1. Cells were coincubated with increasing concentrations of CD47 antagonist and 100 nM fluorescent mouse (A) or human (B) SIRPα tetramers for 1 h at 4 °C, washed, and evaluated by FACS.
