(A) A549 and NCI-H1299 cells were treated with various concentrations of LCA for 24 h, and cell extracts were analyzed to determine the changes of protein expression by western blot analysis. *P < 0.05 and **P < 0.01. The blots were run under the same conditions and the full-length blots were shown in supplementary materials. (B) A549 cells were cultured in 10 μM LCA for 24 h with or without CQ pretreatment (5 μM, 1 h). Cell extracts were analyzed for protein expression using western blot analysis. *P < 0.05 and **P < 0.01. (C) After transient transfection with scramble, ATG7, or ATG5 siRNA for 24 h, A549 cells were treated with 10 μM LCA for 24 h. The indicated protein expression was evaluated by western blot analysis. *P < 0.05 and **P < 0.01. (D) A549 cells were transiently transfected with GFP-LC3 plasmid for 24 h and ATG7 siRNA for 24 h. LCA (10 μM) was added into the cells, and cultured for another 24 h. GFP-LC3 punta were examined using the confocal microscope.
