(a), Backscatter SEM on Dmp1 long bones showed defects in mineralization and more cuboid and round-shaped osteocyte lacuna. Scl-Ab treatment has both improved bone mineralization as well as osteocyte morphologies in WT and Dmp1 null mice. (b),Trichrome goldner staining exhibited more red-labelled osteoid area in Dmp1 bone, which is rescued by Scl-Ab. (c), Quantification data showed significantly improved mineralized bone area ratio according to bioquant (*, p<0.05; n=4). (d), 3-point bending exhibited better mechanical properties in both WT and Dmp1 null mice after antibody treatment (**, p<0.01; *, p<0.05; n=6). (e) The double labeling assay showed no apparent difference in the bone formation rate between the Scl-ab treated and non-treated specimens in the WT group, although the Scl-Ab treated KO group showed a sharp reduction in dye diffusion areas compared to the non-treated KO mice.
