Effect of miR-155 mimic and rosiglitazone on expression of adipogenic and lipogenic markers and lipid accumulation in 3T3–L1 adipocytes. 3T3–L1 adipocytes were incubated with or without rosiglitazone (0.1 μM, 24 h) followed by a 24-h transfection with miR-155 mimic. A, The mRNA levels of Cebpa, Cebpb, Pparg, aP2, adiponectin, and Pgc1a were quantified through qPCR. B, The mRNA levels of Fasn, Acc, Srebp1c, and Scd1 were quantified through qPCR. C, 3T3–L1 adipocytes were transfected with miR-155 mimic. Twenty-four hours after transfection, intracellular lipid was stained with Oil Red O. D, the stained Oil Red O was extracted with isopropanol. The absorbance of the extracted Oil Red O was spectrophotometrically determined at 492 nm after solubilization in isopropanol, and normalized to protein concentrations. The data are expressed as relative expression ratios of at least three independent experiments. The values are presented as the means ± SEM. **, P < .01 compared with control.
