Hyperthermia-induced ERK activity has anti-proliferative effects in mutant KRAS CRC cells. (A,B) Cells were exposed for 16 h to 37 °C or 42 °C, and mock treatment (M), AZD6244 at 0.25, 0.5 and 1.0 μM, or FR180204 (FR) at 25 and 50 μM. Total cell lysates were analyzed by Western blotting, and probed for total ERK1/2, phosphorylated (p) ERK1/2 and ACTIN as described in Figure 2; (C,D) Clonal growth assays as described in Figure 1 were performed with cells exposed to AZD6244 (Ei) at 0.25, 0.5 and 1.0 μM.
